Christina Battista and Zackary Kenz, DILIsym Services Inc., Quantitative Systems Toxicology: Mathematical Modeling Applications in DILIsym

The pharmaceutical industry has increasingly embraced mathematical modeling as a method to predict drug-body interactions. In particular, issues of drug toxicity and drug-drug interactions impact the drug development and approval process. Initially begun as a private-public partnership, DILIsym Services is the leading software platform to help inform issues relating to drug-induced liver injury (DILI) by means of mechanistic mathematical modeling. We will discuss submodels that encapsulate the biochemical processes involved in liver function, which provide a framework for elucidating the mechanisms that may contribute to toxicity. We will walk through some examples utilizing the software to predict hepatotoxicity and infer magnitude of injury from clinically measured biomarkers. There will also be ample time at the end to discuss the process of applying to, working in, and publishing in industry.
